Purpose and use
The antisera are designed to determine the serotypes of Neisseria meningitidis of groups A, B, C, X and Y. These groups include over 90% of N. meningitidis serotypes isolated from patients with severe meningococcal disease (meningitis, sepsis).
Reaction
- Dispense one drop each of antiserum A, B, C, X and Y and a drop of saline solution onto a slide.
- Resuspend a colony of the tested Neisseria meningitidis strain in each drop of antiserum (saline) (use 16–18 hours old cultures for max. 3rd passage).
- Rock the slide in a circular motion for 1–3 minutes.
Reading the results and evaluating the reaction
Take readings against a dark background:
- formation of large agglutinates with distinctive clearing of surrounding fluid: +++
- formation of small clots with slightly turbid surrounding fluid: ++
- formation of very small bordered or thread-like agglutinates with turbid surrounding fluid: +
- no agglutinate formation and the suspension remains unchanged and turbid: -
Evaluation
1. Positive reaction with one antiserum: N. meningitidis type A, B, C, X or Y.
2. Positive reactions with multiple antisera: Polyagglutinable (PA) N. meningitidis.
3. Negative reactions with all antisera: Spontaneously agglutinating (SA) N. meningitidis.
4. Negative reactions with all antisera and saline: Non-agglutinating (NA) N. meningitidis – necessary to indicate that antisera A, B, C, X and Y were used.
Recommendations
The following control strains are recommended for quality control of diagnostic antisera:
- N. meningitidis CCM 4634 – serotype A
- N. meningitidis CCM 4635 – serotype B
- N. meningitidis CCM 4636 – serotype C
- N. meningitidis CCM 4637 – serotype X
- N. meningitidis CCM 4638 – serotype Y

- Antisera are supplied in liquid doses of 1 or 2 ml and are intended for in vitro use only.
- Keep the antisera in a dark place at 2–8 °C and use by the expiry date on the package.
- If the primary packaging of the IVD is damaged, do not use and contact the manufacturer.
- Dispose of unused IVDs and empty packaging in accordance with the Waste Act No. 185/2001 Coll.
Note: Meningococcal strain – interest in sending all strains of N. meningitidis isolated from patients with severe meningococcal disease, or from their contacts, as well as non-agglutinating strains with antisera A, B, C, X and Y.
