Purpose and usage
OXIDASE diagnostic discs are designed for the rapid presumptive detection of cytochrome oxidase. The test differentiates e.g. pseudomonads, aeromonads etc. from gram negative enteric rods of the family Enterobacteriaceae.
Principle
The principle is the detection of the cytochrome oxidase by the color reaction of ethyloxethylparaphenylenediamine with alpha-naphthol during which indophenol blue develops.
Technique
Put the tested culture on the surface of the disc in shape of a line. Read the color reaction during 0.5 - 1 min.
Evaluation
If the reaction is positive (Oxidase +) the inoculated line on the disc turns into deep blue to black. If the reaction is negative (Oxidase -) the inoculated line will stay colorless.
Warning
- OXIDASE discs are designed for laboratory use only.
- Discs should be stored in a dark place at +4°C and should not be used beyond the expiration date shown on the packing.
