Significance and use
The diagnostic disks ITEST UREA are intended for the detection of the enzyme urease. The test allows, for example, the differentiation of the genera Yersinia, Proteus, and Providencia from other gram-negative intestinal rods.
Test principle
The principle is the detection of ammonia, which is produced during the hydrolysis of urea by urease.
Performing the test
Prepare a dense suspension from the tested strain in a test tube with 0.4 ml of sterile distilled water (3rd degree of McFarland turbidity scale). Place the ITEST UREA disk into the test tube with the suspension and incubate at 37 ± 1°C for three hours.
Evaluation
In the case of a positive reaction (UREA +), the suspension with the disk will turn pink-violet. In the case of a negative reaction (UREA -), the suspension with the submerged disk will turn yellow.
Warnings
- The ITEST UREA disks are supplied in packages of 50 pcs (cat. no. DD 511) and are intended for in vitro use only.
- Store the disks in the dark at 2 – 8°C and use them by the expiration date indicated on the label.
- Do not use if the primary packaging of the IVD is damaged and contact the manufacturer.
- Dispose of unused IVD and empty packaging in accordance with the Waste Act No. 185/2001 Sb.
