Purpose and use
ITEST TRIBUTYRIN diagnostic discs are designed for the detection of tributyrin hydrolysis. This test allows easy differentiation of some bacterial species, e.g. Branhamella from Neisseria.
Principle
Tributyrin hydrolysis is reflected by a change in the colour of the indicator.
Procedure
Prepare a thick suspension of the tested strain in 1 ml of buffered saline (pH 7.2) (turbidity value of 3 on McFarland scale) and add 1 ITEST TRIBUTYRIN disc. Incubate at 37 ± 1 °C. The colour reaction will begin to appear after a few hours; make a final evaluation after about 20 hours.
Evaluation
If positive (Tributyrin+), the suspension turns yellow; if negative (Tributyrin-) the colour of the suspension remains red.
Recommendations
It is recommended to use Moraxella (B.) catarrhalis CCM 4391 strain as a positive control and Neisseria subflava CCM 3482 strain as a negative control for quality control of ITEST ONPG diagnostic discs. Both strains are available from the Czech Collection of Microorganisms, Masaryk University, Tvrdého 14, 602 00 Brno.
Important
- ITEST TRIBUTYRIN discs are supplied in packs of 50 (Cat. No. DD 504) and are intended for in vitro use only.
- Keep the discs in a dark place at 2–8 °C and use by the expiry date on the label.
- If the primary packaging of the IVD is damaged, do not use and contact the manufacturer.
- Dispose of unused IVDs and empty packaging in accordance with the Waste Act No. 185/2001 Coll.
