Purpose and usage
BACITRACIN H diagnostic discs are used for presumptive identification of Haemophili in primary culture.
Principle
The test is based on higher resistance of Haemophili to bacitracin (10 i.u.) in comparison with other microbes, joined with the ability of Haemophili to grow in the diffusioin zone of growth factors produced by Staphylococci.
Procedure
Inoculate blood agar with a throat swab or other clinical material and smear as usual. Apply a staphylococcal line onto the inoculated material. Place a sterile ITEST BACITRACIN H disc in it and incubate for 24 hours at 37 °C.
Evaluation
Bacterial colonies which grow after incubation in the diffusion zone of bacitracin in the vicinity of the staphylococcal line can be considered as Haemophili.
